Lycopene Purification with DMSO anti-solvent: Optimization using Box-Behnken’s experimental design and evaluation of the synergic effect between lycopene and Ammi visnaga.L essential oil

Abstract:

Lycopene is an efficient bioactive compound. The current study focuses on its extraction and purification from tomato paste, using a novel efficient approach. We applied the recrystallization method using DMSO as an anti-solvent, which allowed to remove the other carotenoids components in tomato paste and isolate lycopene as crystals. The purity of these crystals has been confirmed by UV–Vis, FTIR and HPLC. We then optimised the purification conditions, by varying the parameters filtrate volume, evaporation temperature and anti-solvent volume, using the Box-Behnken design. The optimal conditions for filtrate volume, evaporation temperature and DMSO volume were 20 mL, 50 °C and 5 mL, respectively, which gave a maximum yield of pure lycopene of 2.73 mg g−1. In a second step, we studied the microbial activity of the obtained lycopene by testing six referenced strains and following the minimum inhibitory concentration (MIC). The MIC values obtained were relatively high, resulting in a low microbial activity. In order to improve it, we combined lycopene with Ammi visnaga L essential oil. The interaction of lycopene with Ammi visnaga led to a synergistic effect on three strains: E. coli, K. pneumonia and S. aureus. These findings revealed the efficiency of DMSO in the purification of lycopene and highlighted the interest of combining lycopene with the essential oil Ammi visnaga L. The approach adopted in this study is original and could be adapted in the industrial field.
