Determination of thiamine and pyridoxine in food supplements by a green ultrasensitive two-dimensional capillary electrophoresis hyphenated with mass spectrometry

Abstract:

An innovative two-dimensional capillary electrophoresis method based on on-line combination of isotachophoretic and zone electrophoretic separation step hyphenated with sensitive and selective mass spectrometry detection was developed, validated and applied for determination of two B vitamins (thiamine and pyridoxine) in various pharmaceutical samples. The developed analytical method was characterized by excellent validation parameters, such as limit of detection (pg·mL−1 levels), linearity (r2 > 0.99), accuracy (recovery in the range of 91.8–101.8%) and precision (intraday relative standard deviation in the range of 1.5–9.5%, interday relative standard deviation in the range of 4.3–10.5%). The developed method was implemented into the quality control of three pharmaceutical formulations containing thiamine and pyridoxine (tablets, effervescent tablets, drops). The determined content of the B vitamins was in very good agreement with the amount declared by the manufacturer. Moreover, the developed method was for the first time investigated according to the criteria of green analytical chemistry. The results obtained from the Analytical GREEnness metric approach and the semi-quantitative Analytical Eco-Scale approach indicate that the method represents an eco-friendly tool for targeted and ultrasensitive quantification of selected B vitamins.
