Amino acid composition of dried cultivated lucerne

Abstract: The amino acid compn. of dried lucerne, cultivated in Bajec, and hydrolyzed in various ways, was detd. by ion-exchange chromatography. After extn. with Et2O and drying at 105° to const. wt., hydrolysis was accomplished with 6N HCl on a water bath under reflux for 40 hrs. or in partially evacuated ampules at 110° for 20 and 24 hrs. The results were higher using hydrolysis in ampules. For the hydrolysis in ampules, 6N HCl + 6N HCO2H (1:1), and an 80fold excess of HI were also used. Higher values for cystine and methionine were obtained by oxidn. with peroxyformic acid before hydrolysis; and, in the case of tyrosine, after hydrolysis with HI. The amino acid compn. varied with different cuttings.
