Some physical properties of tobacco-mosaic virus purified by the combination of chemical and sedimentation methods

Abstract: By the method of diffused light and electron microscope, the weighted av. of mol. wt., the length, and, indirectly, the thickness of tobacco-mosaic virus (I) purified by a combination of chem. and sedimentation methods was studied. During 18 days the dissymmetry and the reducible intensity at 90° angle of the I solns. in distd. H2O and at pH close to neutral point were detd. I was stable in this medium. In borate buffer soln. at pH 9.9, there was on the first day a rapid cleavage of the unstable I particles due to the effect of OH-, but in the following 6 days the remaining I particles were resistant to the further effect of OH-. After this period there was a cleavage even of the stable I particles.
