Determination of sugars by paper chromatography

Abstract: Some modifications of the photometric detn. of d-glucose, d-mannose, and d-xylose on paper chromatograms were studied. Placing solns. on the paper is more exact and rapid with a special 0.1 ml. graduated micropipet than with a mech. pipet. For detection, add phthalic acid directly to 4:1:5 BuOH-AcOH-H₂O, and then apply a soln. of PhNH₂ in Et₂O. Placing solns. into the lines, the length of which is proportional to sugar concn., and photometric evaluation vertically in the direction of the sepn., give for a large range of concns. a linear relation between the color intensity and the amt. of sugar.
